World’s First Human Trial Of A Drug That Regrows Adult Teeth

Most people assume that losing an adult tooth means living without it forever. A team in Japan is working to change that, and they have now begun the world’s first human trial of a drug designed to make adults grow a third set of teeth. Beneath every person’s gums are dormant tooth buds, the biological foundation of a third generation of teeth that the body suppresses and never uses. Researcher Katsu Takahashi at the Medical Research Institute of Kitano Hospital in Osaka spent nearly 30 years studying why those buds stay dormant. His team found that a protein called USAG-1 is the key suppressor, and when they blocked it in mice and ferrets, the animals grew complete new teeth with no serious side effects. In October 2024, the team administered the drug to human volunteers at Kyoto University Hospital, entering Phase I clinical trials.

The trial involves 30 adult males aged 30 to 64 who are each missing at least one tooth, with safety as the primary goal for this first phase. Takahashi said there is a genuine chance some participants could regrow a tooth as a secondary outcome, describing that prospect as one that would make him over the moon. Japan has particular urgency behind this work: more than 90 percent of people aged 75 and older in the country are missing at least one tooth. The team is targeting commercial availability as early as 2030, beginning with children born missing six or more teeth, before expanding to adults with acquired tooth loss. Takahashi described the technology as completely new and said restoring a person’s own natural teeth carries real advantages over implants and dentures. The USAG-1 protein his team targets shares a 97 percent similarity across species, which is why the animal results were so encouraging and gave the researchers the confidence to bring this to people.
